對使用(yòng) STING (E9X7F) Rabbit mAb(上圖,紅色)标記以及使用(yòng) CD45R/B220 (RA3-6B2) Rat mAb (FITC Conjugate) #34399(下圖,綠色)和 DAPI #4083(下圖,藍色)共标記的固定冷凍的小(xiǎo)鼠脾髒組織在低放大倍率(左圖)和高放大倍率(右圖)進行共聚焦免疫熒光分(fēn)析。
對使用(yòng) STING (E9X7F) Rabbit mAb(左圖,紅色)标記的固定冷凍的小(xiǎo)鼠肺髒組織進行共聚焦免疫熒光分(fēn)析。然後将遊離的二抗結合位點用(yòng) Rabbit (DA1E) mAb IgG XP® Isotype Control #3900 封閉,之後用(yòng) COL1A1 (E8F4L) XP® Rabbit mAb (Alexa Fluor® 647 Conjugate) #72827(右圖,僞彩青色)和 DAPI #4083(右圖,藍色)進行共标記。

使用(yòng) STING (E9X7F) Rabbit mAb(綠色)、β-Actin (8H10D10) Mouse mAb #3700(紅色)和 DAPI #4083(藍色)對 MOLP-8 細胞(左圖,陽性)和 A549 細胞(右圖,陰性)進行共聚焦免疫熒光分(fēn)析。
使用(yòng) STING (E9X7F) Rabbit mAb(實線(xiàn))或濃度匹配的 Rabbit (DA1E) mAb IgG XP® Isotype Control #3900(虛線(xiàn))對 A549 細胞(藍色,陰性)和 THP-1 細胞(綠色,陽性)進行流式細胞分(fēn)析。Anti-rabbit IgG (H+L)、F(ab')2 Fragment (Alexa Fluor® 488 Conjugate) #4412 作(zuò)為(wèi)二抗。

幹擾素基因刺激蛋白 (STING, TMEM173, MITA) 是一種跨膜接頭蛋白,它是細胞病原細胞漿 DNA 的天然免疫應答(dá)的一個關鍵組分(fēn) 【1,2】。STING 是一種被發現主要在内質(zhì)網中(zhōng)普遍表達的蛋白 【1】。在細胞漿 DNA 的刺激下,酶 cGAMP 合成酶 (cGAS) 會産(chǎn)生第二信使環-GMP-AMP (cGAMP) 【3,4】。cGAMP 結合并激活 STING 【3,4】。此外,通過 DDX41 和 IFI16 等核酸感應蛋白檢測細胞漿 DNA 會導緻 STING 激活 【5,6】。激活後,STING 與 TBK1 轉位至核周内體(tǐ),并在 Ser366(小(xiǎo)鼠内為(wèi) Ser365)位點被 ULK1 磷酸化【7, 8】。TBK1 激酶磷酸化并激活 IRF-3 和 NF-κB,從而誘導 I 型幹擾素和其他(tā)免疫應答(dá)基因 【1,2,7】。
